3D-printed high-throughput platform to screen antimicrobial peptides for biomaterial surfaces functionalization
Source abstract
Abstract
Antimicrobial peptides (AMPs) are applied to functionalize biomaterial surfaces for infection prevention. However, surface immobilization alters their antimicrobial activity and biocompatibility versus solution states, creating unpredictability. Efficiently screening AMPs that retain both antimicrobial and biocompatible properties on biomaterial surfaces is thus a major challenge. This study developed a 3D-printed high-throughput platform using titanium-based implants to classify 11 AMPs. By combining nonlinear fitting based on neural networks, we revealed that some AMPs exhibited changes in antimicrobial activity and biocompatibility upon surface immobilization. For example, AMP-1 showed the highest antimicrobial activity in solution but lost this functionality when grafted on surfaces. We categorized AMPs into three types: non-antimicrobial, antimicrobial cytotoxic, and antimicrobial biocompatible. The latter group exhibited an optimal grafting density balancing antimicrobial efficacy and biocompatibility. Additionally, preparation parameters from platform regions enabled uniform functionalized surfaces, validating screening effectiveness. This work establishes a high-throughput strategy to identify AMPs suitable for surface functionalization.
