Challenges and opportunities for lysosome-targeting chimeras
Source abstract
Abstract
Targeted protein degradation (TPD) technology has revolutionized therapeutic strategies by hijacking natural cellular degradation systems—particularly the ubiquitin-proteasome and lysosome pathways—to remove specific disease-associated proteins. Emerging as a transformative paradigm within TPD, lysosome-targeting chimeras (LYTACs) transcend the limitations of proteolysis-targeting chimeras (PROTACs) confined to intracellular proteins, enabling precise degradation of membrane and extracellular proteins, thereby broadening therapeutic avenues in cancer and other diseases. This perspective delineates the evolution of TPD technologies, with a focus on LYTACs' groundbreaking advancements—from deciphering the biology of endocytic receptors to pioneering multimodal molecular designs (antibody, fusion protein, aptamer, small molecules, peptides, and nanoparticles). Here, we will discuss and highlight the challenges and translational potentials of LYTACs, providing a primer for researchers interested in TPD.
