Application of pH and hyaluronidase dual-responsive mesoporous carbon nitride nano-drug delivery system for chemodynamic therapy and chemotherapy combination therapy of non-small cell lung cancer
Source abstract
Abstract
To date chemotherapy remains the primary treatment for non-small cell lung cancer (NSCLC) across various stages. However, chemotherapy drugs have many limitations, including low water solubility, inadequate targeting, and remarkable side effects, which contribute to the diminished effectiveness of chemotherapy. A hyaluronic acid (HA)–modified mesoporous carbon nitride–copper oxide nano-drug delivery system (PCN[sbnd]CuO[sbnd]HA@AZD) loaded with Osimertinib (AZD) has been developed for chemotherapy/chemodynamic combination therapy of NSCLC. Given its high photostability, good biocompatibility, and low systemic toxicity, mesoporous carbon nitride was used for loading CuO and AZD. The released copper(II) ions could generate ٠OH by reacting with H2O2 through Fenton reaction, triggering the apoptosis of the tumor cells, and achieving chemodynamic therapy (CDT). To avoid premature release of chemotherapy drugs, HA was modified on the surface of the nano-drug delivery system as the gated structures that could achieve dual stimulation responsiveness of pH and hyaluronidase (HAase). The as-synthesized PCN[sbnd]CuO[sbnd]HA@AZD has pH/HAase dual responsiveness and good efficacy in vivo and in vitro with negligible systemic toxicity using chemotherapy and CDT combination therapy. The PCN[sbnd]CuO[sbnd]HA@AZD provides a simple and effective therapeutic method for NSCLC and shows a great potential in biomedical applications. © 2024
