Synthesis, aggregation and drug solubilization of block polyethers: Influence of branch number and central aromatic group
Source abstract
Abstract
Abstract Three branched PEO-PPO polyethers TEPA, PEI and PAR with identical PEO and PPO blocks but varying branch numbers were synthesized. Commercially available tetrabranched Tetronic T904 was chosen for contrast. The influence of branch number and central aromatic group on self-assembly behaviors of four polyethers were investigated. The solubilization of hydrophobic drug olmesartan medoxomil and in vitro drug release behavior were evaluated for aggregates accordingly. The order of branch quantity is PEI > TEPA>T904, and they all formed micelles. The smaller cloud point, higher critical aggregation concentration (CAC), lower micropolarity, larger micellar size, higher drug solubilizing ability and slower drug release rate were characteristics of hyperbranched polyether PEI. The introduction of three aromatic rings to hyperbranched polyether PAR also dramatically affected the property of polyethers, and PAR spontaneously assembled into vesicles. PAR displayed the best solubilization capacity and prominent sustained drug release performance, manifesting promising application of PAR in drug delivery systems.
