Nanomedicine-Driven Modulation of the Microbiota for Cancer Therapy
Source abstract
Abstract
Microbiota are composed of a vast number of commensal microbes predominantly residing in the gastrointestinal tract. The microbiota play a critical role in maintaining host health through intricate interactions with the immune system. This symbiotic relationship begins early in life, establishing a stable microbial composition that supports immune homeostasis. However, disruptions in the balance of these microbial communities, known as dysbiosis, and alterations in their metabolic outputs have been linked to various diseases, particularly the initiation and progression of cancer. Cancer is increasingly recognized as a disease with both genetic and chronic inflammatory underpinnings, driven in part by the tumor’s ability to evade host immune surveillance. The microbiota, through its dynamic crosstalk with the immune system, can significantly influence the tumor microenvironment. Modulating microbial composition and metabolites offers a promising strategy to reprogram the immunosuppressive and pro-tumoral microenvironment into one that promotes antitumor immunity. It can also be combined with clinical cancer therapies to further enhance antitumor efficacy. Nevertheless, the interplay among microbiota, immune responses, and cancer is highly complex. Effective antitumor strategies require precise modulation of microbial communities to activate specific biological pathways and target defined spatial locations. Nanotechnology presents a cutting-edge approach to achieving such specificity. Nanotechnology can modulate the microbiota and its metabolites, allowing treatments to target certain tissues or cells directly. These nanoscale systems can be engineered to respond to unique enzymatic or physiological conditions within the tumor, lymphoid, or gastrointestinal microenvironment. The application of nanotechnology to shape the microbiota and enhance host antitumor immunity holds transformative potential. As a standalone therapy or in combination with other treatments, nanomedicine-driven modulation of the microbiota represents a promising frontier in cancer therapeutics. It helps to address critical unmet needs in the clinical and pharmaceutical fields.
