Abstract

The rapid systemic clearance of conventional nanomaterials significantly limits their retention and therapeutic efficiency, especially in cancer treatment. Prostate cancer, known for its resistance to conventional therapies, demands innovative approaches for effective management. In this study, we developed a multifunctional nanoplatform, SFe@PDA-BD MIPs, to overcome these challenges. The nanoplatform features a porous Fe 3 O 4 core loaded with the drugs doxorubicin (DOX) and enzalutamide (ENZ), coated with polydopamine imprinted with human serum albumin (HSA). This unique molecular design facilitates the in situ adsorption of HSA, forming a protein corona that extends circulation time and enhances immune evasion. Upon near-infrared (NIR) light irradiation, the nanoplatform generates localized hyperthermia and reactive oxygen species (ROS) through a Fenton-like reaction mediated by the Fe 3 O 4 core, thereby enhancing drug release in the acidic tumor microenvironment. The combined effects of androgen receptor inhibition by ENZ and chemotherapy with DOX lead to effective tumor regression while demonstrating significant anticancer potential without systemic toxicity. Thus, SFe@PDA-BD MIPs provide an innovative approach for targeted prostate cancer treatment, integrating photothermal therapy, chemotherapy, and biomimetic camouflage. This multifunctional strategy addresses key limitations of conventional nanomaterials, offering a promising avenue to enhance therapeutic efficacy while ensuring systemic safety.
