Blinking Nanoparticles as Super-Resolution Bioimaging Probes: A Review
Source abstract
Abstract
Super-resolution microscopy has revolutionized the visualization of nanoscale subcellular structures via bypassing the diffraction limit and has become a powerful tool in biological research. Among various super-resolution techniques, single-molecule localization microscopy (SMLM) offers exceptionally high resolution in the range of 10–100 nm. This review presents a concise overview of the fundamental concepts of SMLM and various types of nanoparticle-based fluorophores that can be utilized for SMLM-based super-resolution bioimaging. First, we summarize the unique characteristics, advantages, and drawbacks of various blinking nanomaterials that are used in SMLM imaging applications. Next, we focus on the applications of these blinking nanomaterials in SMLM-based super-resolution imaging of various biological structures and processes. Finally, we discuss the challenges associated with nanoparticle-based SMLM probes and their potential solutions, providing insights into the advancement of this cutting-edge imaging modality.
