Abstract

Circulating tumor cells (CTCs) are cells that have detached from primary tumors and entered into peripheral blood; they are often associated with cancer metastasis. Accurate and sensitive detection of CTCs is critically important for cancer diagnosis. In this work, we developed a microfluidic electrochemical system for detecting and releasing CTCs (μFEC-CTCs system). Screen-printed gold electrode (SPGE) was first prepared by modifying the carbon electrode surface with gold nanoparticles and then decorating a tetrahedron DNA framework with 17 base pairs on SPGE as a capture probe. The microfluidic chip was prepared by bonding the nanomaterial decorated electrode with a PDMS layer of “serpentine” shaped microchannels. We connected the microfluidic chip with a syringe pump and electrochemical workstation to complete the system construction. The μFEC-CTC system that we developed has excellent detection performance for CTCs with a limit of detection (LOD) of 10 cells in both buffer and mimic blood samples. The release efficiency reached 80% and the survival rate of the released cells was as high as 90%. The as-prepared μFEC-CTCs system offers a promising tool for the detection of CTCs, which could greatly contribute to the early clinical detection of cancer.
