Bioinspired nanoplatform for catalytic-photothermal synergistic antibacterial therapy and accelerated diabetic wound healing
Source abstract
Abstract
The hyperglycemic microenvironment of diabetic wounds is highly susceptible to bacterial infections and biofilm formation. This, coupled with inflammation dysregulation and impaired angiogenesis, poses a significant challenge to effective clinical management. Mild photothermal therapy is considered a promising anti-infective strategy for diabetic wounds owing to its excellent biocompatibility and pro-angiogenic properties. However, the therapeutic efficacy of photothermal therapy alone remains insufficient for the complex microenvironment of diabetic wounds. To address this limitation, we constructed a multifunctional nanoplatform (MMD) for synergistic therapy. This platform employs three-dimensional MoS 2 nanoflowers as a carrier, onto which Mn-NC nanozymes with glucose oxidase-like activity are deposited. The composite is further coated with dextran for enhanced biofilm localization and retention, forming an integrated therapeutic system. In the diabetic wound microenvironment, MMD orchestrates a self-amplifying antibacterial loop: Mn-NC consumes endogenous glucose to produce H 2 O 2 , while MoS 2 provides mild photothermal effect under NIR irradiation. This concerted, fuel-triggered dual action achieves robust bacterial killing without the thermal collateral damage inherent to conventional photothermal therapy. In vitro evaluations revealed that this multimodal synergistic approach achieved high antibacterial efficacy, eliminating over 99% of both E. coli and S. aureus and effectively clearing associated biofilms. In vivo studies revealed that MMD + NIR treatment significantly cleared wound bacteria, accelerated inflammation resolution, and promoted re-epithelialization, collagen deposition, and angiogenesis. Consequently, MMD + NIR treatment achieved a high-quality healing ratio of 96.24 ± 1.36% in a diabetic rat model with infected wounds. An integrated nanomaterial-based approach with therapeutic synergy is proposed in this study for diabetic wound infection, offering dual functionality for precise antibacterial action and active promotion of healing.
